From: Control of replication initiation by the Sum1/Rfm1/Hst1 histone deacetylase
hst1Δ and rfm1Δ showed synthetic lethality with orc2-1. An orc2-1 hst1Δ (AEY3941) and an orc2-1 rfm1Δ (AEY3940) strain carrying pURA3-ORC2 (pAE1316) were tested on 5-FOA medium for their ability to lose the ORC2 plasmid when additionally provided with a LEU2-marked ORC2 plasmid (pAE1315) or empty LEU2-vector (pRS315). Strains were incubated for three days at 30°C.
