Effect of mutating the Ets sites within the LAT promoter. Site-directed mutagenesis was used to mutate the Ets site centered at -360 (mutEts -360), the Ets site centered at -390 (mutEts -390), or both Ets sites (mutEts -360/-390). Mutations were introduced into the -1000 to -1 promoter fragment within the pGL3-basic vector. Specific base pair changes are shown in Fig. 1. Reporter plasmids were transiently transfected into Jurkat T cells and fold activation was determined as previously described. The results depict the average fold induction and s.e.m. from three or more independent experiments.
