Chelation of intracellular Ca2+attenuates expression of Egr-1 in thrombin or carbachol stimulated 39M1-81 fibroblasts and in 1321N1 glioma cells. 39M1-81 (A) or 1321N1 (B) cells were preincubated for 30 min with the calcium chelator BAPTA-AM (50 μM). Cells were stimulated with either thrombin or carbachol for 1 hour. Nuclear extracts were prepared and subjected to Western blot analysis. The blots were incubated with an antibody directed against Egr-1.
