Interaction of USF1 and USF2 transcription factors with the AβH-J-J P1 promoter. (A) EMSA were carried out on 2 μg of Hela nuclear extracts as described in the Experimental procedures using P1/USFmer probe (Table 1). Probe was incubated with nuclear extracts in the absence of competing oligonucleotides (lane 1) or in the presence of the indicated competing oligonucleotides (lanes 2–5). Arrows and asterisk indicate the specific and non-specific complexes respectively. (B) Supershift assay was performed using P1/USFmer probe and 2 μg of nuclear extract from HeLa cells; probe was incubated with nuclear exctract in the absence of antibody (lane 6) or in the presence of antibodies against USF1 factor, USF2 factor or both (lanes 7, 8, and 9 respectively).
