ISWI is acetylated in the HAND domain. (A) Schematic representation of ISWI, the extent of known domains (first line) and of deletion mutants used for mapping the acetylation site. Numbers correspond to the first and the last amino acids of the proteins. They were expressed in E. coli with either HIS6- or glutathione-S-transferase- (GST-) tags. (B) The different ISWI fragments displayed in (A) were acetylated in vitro with GCN5. Reactions were resolved by SDS-PAGE and visualized by autoradiography (upper panel) and Coomassie staining (lower panel). Black dots label the positions of the ISWI fragments. An asterisk marks the band corresponding to GCN5.
