From: Interleukin-4 induction of the CC chemokine TARC (CCL17) in murine macrophages is mediated by multiple STAT6 sites in the TARC gene promoter

Figure 5

Probes containing the mTARC putative STAT6 sites T1, T2, T4 and T11 bind stable STAT6 complexes in vitro. Nuclear extracts were prepared from C57BL/6 peritoneal BIOgel-elicited Mθ treated +/- 20 ng/ml recombinant murine IL-4 or TFN-γ for 1 hour. Extracts were incubated with radiolabelled mTARC T1-11 putative STAT6 site probes (data not shown for T3 and T5-T10, which did not bind STAT6) for 15 minutes before EMSA was performed (A). Specificity of binding was demonstrated by pre-incubating nuclear extracts +/- specific STAT Abs or excess cold competitor mTARC T1 or T4 putative STAT6 site probes (B). Bands labelled * represent nonspecific DNA-protein interactions, FP shows the top of the free probe and -Ori denotes the origin of the gel.
