mTARC mRNA and protein are strongly IL-4-induced and display temporal congruence. C57BL/6 peritoneal BIOgel-elicited Mθ were cultured in OptiMEM alone (untreated) or in the presence of 20 ng/ml recombinant murine IL-4 or EFN-γ or 40 ng/ml LPS. A, Total RNA was harvested at the designated time points and subjected to real-time PCR. Relative mTARC cDNA copy number was interpolated from an internal standard curve and expressed as a ratio of mTARC/HPRT +/- SEM, the results shown are representative of more than 3 similar experiments. B, Supernatants were collected at same time points for protein determination by ELISA. Protein results represent data from duplicate samples +/- SD.
