The C-terminal fragment of Cdc23 S424 ::Tcs is not localized to the nucleus after cleavage by TEV protease. A. cdc23 S424 ::Tcs strain (P1323) was grown in plus (-TEV) or minus thiamine (+TEV) media at 25°C for 24 h, then shifted to 37°C for 4 h before fixation with methanol and acetone.
