Localization of the -65 DNase I hypersensitive site upstream of XIST. A. Using an Xa hybrid to limit background from cross-hybridization to human material, two additional restriction digests were performed on different DNA preparations after increasing DNase I treatments to refine the location of the -65 site. The Bgl II fragment is 9.3 kb and the fragment resulting from HS and restriction digestion is 5 kb (grey arrow, left panel). The Sca I fragment is 11.8 kb and the fragment resulting from HS and restriction digestion is 8 kb (grey arrow, right panel).
