PAT relaxes positively supercoiled DNA as well as negatively supercoiled DNA. In lane 1 is positively supercoiled pUC18 (S1), lane 2 contains negatively supercoiled pUC18 (S2), lane 3 contains positively supercoiled DNA relaxed, lane 4 contains negatively supercoiled DNA relaxed. Lanes 3 and 4 contain 110 ng of supercoiled DNA, 9 ng PAT in the topoisomerase buffer with 2.5 mM MgCl2.
