  2. a. The abbreviation lacIZ33 is used to denote the triple mutant Ï•(lacI33-lacZ) bearing the lacIQpromoter-up mutation in the lacI promoter, a 212-bp deletion fusing lacI to lacZ, and a +1 frameshift mutation in the lacI sequences near the fusion junction [13].
  3. b. Strains designated "red" bear the phage λ red recombination genes, which replace the recC-ptr-recB-recD gene cluster in the E. coli chromosome.
  4. c. Primers used in polymerase chain reactions to generate antibiotic cassettes for gene replacements are described in Table 3.
  5. d. Strains designated "(lacIZ33Y)2-" bear a duplication of lacIZ33 and lacY, with the genetic element listed after the hyphen (tet-oriR6Kgamma or cat) inserted between the two copies.
